Bacteria Collections Lab
Purpose: The purpose is to figure out where a lot of bacteria are and how bacteria look like (different shapes, colors,..)
Material: petri dish with agar, q-tip, crystal violet, iodine, 95% ethanol, safranin, inoculating loop, slide, destilized water, slide, microscope, Bunsen burner
Method:
1. Divide the petri dish into 4 quadrants.
2. Swipe 4 different (dirty) surfaces with the q-tip and gently place it on the 4 quadrants.
3. Let the bacteria grow for days.
4. Sterilize inoculating loop with the Bunsen burner
5. Clean the slide with water & Kim wipe
6. Smear with the inoculating loop bacteria as thin as possible onto the clean slide.
7. Sterilize inoculating loop again.
8. Gently burn the bacteria on the slide by passing through the flame of the Bunsen burner (~2 – 4x)
9. Apply Crystal Violet on the bacteria and rinse with destilized water after ~40sec.
10. Apply Gram’s Iodine (IKO) on the bacteria and rinse after ~1min.
11. Apply 95% ethanol on the bacteria for ~5sec and rinse immediately
12. Apply Safrinin for ~1min on bacteria and rinse with destilized water. Blot dry.
13. Prepare 4 different slides like that.
14. Disinfect your work area.

14. Disinfect your work area.

- Colony Color: cream colored yellowish; some orange dots; some brown dots
- Colony Texture: between translucent and opaque
- Colony Surface: shiny
- Colony Margin: irregular, feathery on the edge, lobate
- Colony Form: irregular
- Colony Elevation: raised
- Gram positive bacteria (dark purple)
- Colony Number: ~3
- Colony Color: cream colored yellowish; some orange dots; some brown dots
- Colony Texture: between translucent and opaque
- Colony Surface: shiny

Conclusion:
This lab proved that things my jacket are surprisingly super dirty and are even worse than for example money!
I should have timed the slides better (probably do more than 1 at a time)
